Welcome back to episode four of “Library Prep with the I.DOT Liquid Handler.” Today, we're talking about the Illumina Nextera XT DNA Library Preparation Kit. If you use this in your lab, you like saving money and time, watch below:
The I.DOT Liquid Handler is designed to dispense microliter scale volumes precisely and accurately without the need for purchasing pipette tips for library prep. The I.DOT Liquid Handler is a non-contact microfluidic dispenser that does not require any tips, training, or automation experience.
The Nextera XT DNA Library Prep Kit streamlines the process with a single enzymatic step for fragmentation and tagging DNA. The I.DOT is designed to dispense up to 65% glycerol, so even the most viscous enzymes can be dispensed using this flexible device. The I.DOT can be used with all reagents from this kit (Table 1).

It also eliminates the need for library quantification before pooling with a bead-based normalization step (Fig. 1).
This allows for faster library preparation with less hands-on time compared to other methods.
When combined with automated liquid handling systems like the I.DOT Liquid Handler, this kit could potentially be the fastest and most hands-off library prep workflow available.
