In the realm of next-generation sequencing (NGS), DNA purification is a crucial step in library preparation. This process eliminates unwanted contaminants, ensuring the purity of the sample for downstream applications. Traditionally, DNA purification has been performed manually, which can be time-consuming, labor-intensive, and prone to errors.
Introducing the G.PURE NGS Clean-up Device, an automated plate washer and liquid dispenser specifically designed for cellular assays and bead-based DNA purification. This innovative device offers several advantages over conventional manual methods, including:
-
Reduced processing time: The G.PURE streamlines the bead-based DNA purification process, significantly reducing the time required from hours to just minutes (Fig. 1).
-
Enhanced consistency: By automating the process, the G.PURE eliminates the potential for human error, ensuring consistent and reproducible results across samples.
-
Miniaturization compatibility: The G.PURE is compatible with miniaturized workflows, allowing for the purification of DNA samples with starting volumes as low as 5 μL.
-
Reduced consumables: The G.PURE's non-contact dispensing and removal of liquids eliminate the need for pipette tips, saving money and reducing waste.
Figure 1. The G.PURE provides a fast and automated workflow for bead-based DNA purification. Standard manual bead cleanup workflows are time-consuming and require many pipettes due to the several removals, wash and dispense cycles used to purify the DNA. The G.PURE performs the critical wash steps 5x faster than conventional purification while requiring no pipetting steps.
To evaluate the G.PURE's performance, we compared its effectiveness to manual bead cleanup using unpurified cDNA post SMART-seq2 library preparation. Bioanalyzer traces (Agilent Biotechnologies) revealed that the G.PURE provided equivalent or better results in DNA purification compared to manual bead cleanup, while taking only a fraction of the time (Fig. 2).
Figure 2. Prior to bead cleanup, the Bioanalyzer (Agilent Biotechnologies) trace of the unpurified cDNA has a dominant peak with smaller fragments (A). The same sample underwent DNA bead cleanup by manual prep, which resulted in a trace with the expected size distribution and most of the smaller fragments removed (B). The sample processed by the G.PURE with one wash step (C) and two wash steps (D) also resulted in ideal traces with minimal small fragments present. The G.PURE also produced similar results in a 1/5th miniaturized sample as with standard volumes (E).
The G.PURE represents a significant advancement in DNA purification technology, offering a faster, more consistent, and cost-effective solution for NGS library preparation. Its compatibility with miniaturized workflows further expands its applications in various research settings.
