Library Prep with the I.DOT Liquid Handler & Illumina TruSeq Nano DNA HT Library Prep Kit

Learn how the I.DOT Liquid Handler streamlines Illumina TruSeq Nano DNA HT library prep, saving time and improving efficiency in labs.

Did you know that liquid handling automation saves labs hours of hands on time? Welcome to episode one of Library Prep with the liquid handler that has the lowest barrier to entry, the I.DOT.

In this episode, we are covering the Illumina TruSeq Nano DNA HT library prep kit, and this one is very special for so many reasons. The reason researchers love working with this kit is because it requires a low sample input, it has excellent coverage, high flexibility, and it has 96 unique dual indexes. Our customers really love the I.DOT Liquid Handler because it enables labs to dispense microliter scale volumes accurately and precisely. Keep reading to learn how.

Following the TruSeq Nano DNA library prep workflow (Fig. 1), right off the bat, you can use the I.DOT to dispense your fragmentation enzyme as well as your magnetic beads for fragmentation clean-up. Researchers can take their library prep automation workflow to another level by using the plate washer, the G.PURE NGS Clean-up Device.

The G.PURE is a tipless plate washer and it can be used for all of your bead-based clean-ups and size selection steps in your library prep process.  It can also dispense your ethanol and resuspension buffer. The combination of the I.DOT and G.PURE bundle can automate 80% of your library prep workflow with little training and no automation experience required.

For adenylation, ligation, and PCR, you simply select the standardized liquid class on the IDOT that matches the viscosity of whatever you're dispensing. So the I.DOT can dispense the A-tailing mix, the ligation buffer, the beads, the stop ligation buffer, the PCR mix, the PCR primer cocktail beads – you get it.

And then, of course, you can use the G.PURE to clean up and dispense ethanol and resuspension buffer as needed.

Finally, you can normalize and pool your libraries using the I.DOT source plates with variable volumes of your library and backfilling with PCR-grade water.

And that's it! High quality libraries in a fraction of the time. Book your personalized demo today!
