Following the TruSeq Nano DNA library prep workflow (Fig. 1), right off the bat, you can use the I.DOT to dispense your fragmentation enzyme as well as your magnetic beads for fragmentation clean-up. Researchers can take their library prep automation workflow to another level by using the plate washer, the G.PURE NGS Clean-up Device.

The G.PURE is a tipless plate washer and it can be used for all of your bead-based clean-ups and size selection steps in your library prep process.  It can also dispense your ethanol and resuspension buffer. The combination of the I.DOT and G.PURE bundle can automate 80% of your library prep workflow with little training and no automation experience required.

For adenylation, ligation, and PCR, you simply select the standardized liquid class on the IDOT that matches the viscosity of whatever you're dispensing. So the I.DOT can dispense the A-tailing mix, the ligation buffer, the beads, the stop ligation buffer, the PCR mix, the PCR primer cocktail beads – you get it. 

And then, of course, you can use the G.PURE to clean up and dispense ethanol and resuspension buffer as needed.

Finally, you can normalize and pool your libraries using the I.DOT source plates with variable volumes of your library and backfilling with PCR-grade water.
